Clarity Oligo-RP  is specially designed for the reversed-phase purification of oligonucleotides, balancing both hydrophobic and polar selectivity. The filler uses organic hybrid TWIN™ technology, which improves the selectivity and column efficiency for analyzing oligonucleotides.

 

 

Clarity Oligo-RP™  used for the purification and characterization of oligos

  • Can separate N-1 failure sequences in target oligonucleotides, with purity reaching over 90%; 
  • Supports purification of DNA, RNA, thiol-modified, and various labeled oligonucleotides after detritylation;
  • Can meet sample loading needs of 5 μmol or more based on preparation specifications and particle size;  
  • Can purify oligonucleotides up to 60 mers in length;  
  • Can be used as a reverse-phase HPLC column for oligonucleotide quality control (QC).

 

 

Prepare and purify on Oligo-RP

Reverse-phase separation of oligonucleotides has more advantages compared to other separation methods like ion exchange. The Oligo-RP stationary phase allows for high sample loading, excellent recovery and purity, and avoids extra purification steps. This is thanks to the ion-pair separation method that separates de-tritylated oligos from failed sequences and other impurities.

 

Separate N-1 failed sequences from the target N sequence

The Oligo-RP filler is specially designed to match the various properties of nucleotides and has a reactivity pattern that fits itself. The filler surface contains hydrophobic, dipole, π-π, and hydrogen bond donor/acceptor sites. This combination of interactions, along with ion-pair reagents, allows for highly selective differentiation between nucleic acids. As a result, this filler can detect extremely subtle differences in nucleotide sequences, like a single base difference (N and N-1) or a single base substitution.
