Separation of impurities in cosmetic ingredient ceramide AP


Ceramide AP is a naturally occurring lipid component in the skin's stratum corneum, belonging to the ceramide family, and mainly involved in building and repairing the skin barrier. It helps maintain healthy skin by strengthening intercellular lipid structures and reducing moisture loss, especially benefiting dry, sensitive, or barrier-damaged skin.

      A user provided a Ceramide AP sample and wants a good analysis of it, requiring that the impurities before and after the main peak are separated by ≥1.5, and that the main peak has a good shape.

Ceramide AP structural formula

 

Ceramide AP is quite fat-soluble. When using methanol as the solvent, a sample concentration of 1 mg/mL and ultrasonic treatment for over 40 minutes can achieve complete dissolution. However, since the autosampler tray is kept at 15°C, continuously injecting samples at 1 mg/mL can lead to gradual precipitation. Therefore, we tried further diluting it to 100 μg/mL with 90% acetonitrile. Tests showed no precipitation, and the analysis results were consistent.

Because ceramide AP has weak UV absorption, we used the versatile nano-scale laser particle detector NQAD for detection. Using a DAISOPAK ODS-P column under a high-organic methanol-water gradient, we got good analytical results for ceramide AP, as shown in Figure 1.The main peak retention time is 14.5 minutes, with an asymmetry factor of 0.97. The peak shape is good, and the resolution between the main peak and the adjacent impurities is greater than 1.5, meeting the user's requirements.。

Mobile phase: A, water; B, methanol B% 95%(0 min) -100%(8 min) -100%(35 min) -95%(35.1 min) -95%(45 min)  

flow rate:1.0 mL/min

Column temperature:40 ℃

Check:NQAD(Evaporator Temperature :60 ℃、Nebulizer Temperature:40 ℃)

Sample volume:50 μL

concentration:100 μg/mL(After dissolving, dilute with 90% acetonitrile)
 

In summary, using a DAISOPAK SP-100-3-ODS-P 4.6 mm i.d. × 250 mm column under a high-organic methanol-water gradient gives good analytical results for ceramide AP, meeting user requirements. However, the ceramide AP sample has poor solubility, so you need to watch out for sample concentration and analysis conditions during the process to avoid precipitation.
