Key Points for Use and Installation
1. Prep before installation: The column comes stored in methanol, so you need to flush it with methanol at a flow rate of 1.0 mL/min for 20 minutes first (don’t connect it to the chromatograph or detector).
2. Connection location: It must be installed between the mixer and the injector, not after the injector, to avoid sample adsorption.
3. Notes on gradient elution:
- It’s recommended to extend the post-run equilibration time a bit.
- If you need faster equilibration, you can choose a smaller column size (like 4.0×30mm).
Notes
Ion pair reagents: Tolerates tetrabutylammonium hydroxide, sodium heptanesulfonate, etc., but may shorten lifespan or cause irreversible adsorption.
Mass spectrometry coupling: Trace leachables may occur; it’s recommended to flush in advance and do a compatibility test.
Cleaning and storage:
- Salt-free systems: Rinse with 80% methanol in water for 20 minutes before storing;
- Salt-containing systems: First rinse with 10% methanol in water, then with 80% methanol in water before storing.
