7 Essential Steps for Effective Peptide Synthesis Using Purepep Chorus

Employ modular design and intuitive software workflows to meet stringent purity requirements.
Pre-programmed standard method —— It can be used directly or as a template for custom methods.
Import sequence —— Easily create synthesis processes using the key library.
Multitasking —— While one synthesis process is running, develop a plan for another synthesis process.
Automatic calculation —— Use the reagent preparation and dosage calculator built into the software.
All container groups can be configured. —— Achieve fast, independent induction heating and real-time UV monitoring.
Flux is upgradeable. —— From 2 reaction vessels to 4-6.
The Simple 7-Step Method: Peptide Synthesis Using Purepep Chorus
The first 1 Step:
Group the reaction vessels according to the principle of synchronous or continuous operation. Different groups can carry out different operational steps.
The first 2 Step:
Input parameters, including coupling method, reagent excess, and resin information, are used for record keeping and calculation.
The first 3 Step:
Add peptides from the library, or create new peptides using the sequence editor. Select the cycle corresponding to the specific chemistry within the group; for adding expensive reagents, use the Single-Shot method that eliminates the need for rinsing. TM 。
The first 4 Step:
Choose whether to use the default positions for amino acids and reagents, or rearrange them as needed.
The first 5 Step:
Assign the pre-programmed or custom chemistry protocol to all cycles in all groups. If performing heating synthesis, select the default temperatures for deprotection and coupling. If any steps, cycles, RVs, or groups have exceptions, make corresponding selections as well. The first 6 Step:
Wait for the software to calculate all required reagent weights and volumes, as well as the theoretical yield and estimated synthesis time.
The 7th Step:
Synthesis can be started from scratch or from any desired cycle and step. During the synthesis run, monitor UV and temperature data in real time, and follow the countdown to complete peptide synthesis.

Intellisynth™ U V Monitoring
You can configure the proprietary Intellisynth real-time UV monitoring system on any number of reaction vessel groups as needed, enabling automated optimization of deprotection reactions. By monitoring the solution during the mixing process, you can precisely control reaction time and the number of repetitions, ensuring that the deprotection reaction proceeds efficiently while avoiding unnecessary waste of time or reagents.
You can view real-time UV data during the composite run, or you can view the complete data for all completed composites.
A single chart shows the reaction progress within a single cycle. The overall chart highlights differences among various cycles in terms of time and repetition count, thereby providing valuable diagnostic information for identifying synthesis failure points in complex sequences.

Induction heating
You can configure induction heating on any number of reaction vessel groups as needed. This equipment can accelerate chemical reactions and enhance the purity of complex reaction sequences. In a set of six reaction vessels, the heating conditions for each vessel can be set independently, enabling temperature scans to optimize processes or accommodate temperature conditions required for parallel synthesis of sequences involving different steps. During a synthesis run, you can view real-time temperature data, as well as complete temperature profiles for all completed syntheses.

21 CFR Part 11 Compliance and IQ/OQ/PO
The PurePep® software was designed with compliance in mind according to 21 CFR Part 11, ensuring that the instrument can be used in a regulated CGMP laboratory environment. We can also provide IQ/OQ support and PQ guidance.
The main software features include:
-
User Management
-
Electronic signature
-
Audit trail
-
Easy-to-view reports enhance traceability.
