Self-Check Guide | Device Restart   Precise Calibration

The Spring Festival holiday is over, and the lab is about to resume operations! During the shutdown period, equipment left idle and changes in the environment could both harbor hidden operational risks. Rushing to restart equipment prematurely can easily lead to equipment malfunctions and inaccurate experimental data.

I. The First Step in Commencement: A Comprehensive Inspection of the Laboratory Environment to Eliminate Basic Hazards

 

 

 

Before resuming work, first conduct a “big-picture” assessment to identify and mitigate safety and equipment operation risks at the source. Focus on performing these three key inspections:

 

1. Safety inspection of water, electricity, and gas: Open the laboratory’s main water valve and main power switch, and check that there are no leaks in the plumbing system, no short circuits or tripped circuit breakers in the electrical circuit, and that all socket connections and equipment wiring are secure. Carefully inspect each high-pressure gas cylinder one by one, ensuring that the pressure-reducing valves are closed and that there is no damage to the piping. Use soapy water to test the PEEK fittings and capillary lines for any leaks. Verify that hydrogen cylinders remain at least 15 meters away from oxidizers. Remove all “Work-Stoppage” signs and keep a detailed record of the removal.

 

2. Verification of reagent and sample conditions: Check that flammable and highly toxic reagents (such as methanol and acetonitrile) stored in the explosion-proof cabinet are properly sealed, with no signs of evaporation or leakage, and that the cabinet temperature is ≤30℃. For highly toxic reagents, verify the inventory records using a two-person, dual-lock system to confirm that there are no storage abnormalities. Inspect the samples stored in the low-temperature refrigerator to ensure that the voltage-stabilized power supply is functioning normally, the temperature fluctuation is ≤±2℃, and the samples show no signs of deterioration. Re-filter any unused mobile phases by ultrasonication, and clearly label all information for easy identification.

 

3. Verification of Waste Liquids and Fire-Fighting Facilities: Check that the waste liquid drums stored separately before the holiday are properly sealed, with no leaks or mixed storage, and that their hazardous waste labels are clearly visible. Confirm that fire extinguishers, eyewash stations, and other fire safety equipment are within their validity periods, unobstructed, and fully functional. Also, ensure that information such as laboratory emergency contacts and fire department phone numbers is clearly displayed.

 

II. Core Component: Step-by-step restart of precision equipment to avoid idle-time losses.

Due to the different maintenance methods required for various precision equipment, restarting them must follow “dedicated procedures.” It’s crucial to focus on thoroughly flushing pipelines, resetting components, and gradually powering up the equipment, thereby avoiding direct startup that could cause damage.

 

(1) Restarting the Liquid Chromatograph (HPLC/UPLC): Proceed step by step, and strictly prevent bubbles and contamination.

1. Pre-boot inspection

Visual inspection: Check the pump head, pipeline connections, and test pool for any signs of leakage; ensure that the column oven is clean and dry.

Power restored: First, connect the main power supply and wait 30 seconds before sequentially turning on the detector, pump, and column oven power supplies to avoid high-current surges.

2. Solvent System Venting

Replace with fresh mobile phase: It is recommended to re-filter and sonicate the mobile phase that has not been fully used before the holiday to prevent bacterial growth or component volatilization.

Remove air bubbles: Open the pump head vent valve, use a syringe to draw in and flush the cleaning solution (1:1 methanol-water) until no air bubbles remain in the pipeline, then close the vent valve.

Gradient System Validation: Run the low-flow-rate gradient program and observe whether the pressure curve is stable. If the fluctuation exceeds 5%, check the check valve or filter cartridge.

Check column storage liquid: For chromatographic columns sealed with pure methanol before the holiday, install them directly (if sealed with buffer salts, first transition using a methanol-water mixture).

Low-pressure start: Set the initial flow rate at 0.2 mL/min, gradually increase it to the experimental flow rate, and after the pressure stabilizes, record the initial value. If the deviation from the pre-test value exceeds 10%, investigate the cause.

Column efficiency verification: Inject the standard sample and observe whether the retention time, theoretical plate number, and tailing factor fall within the acceptable ranges.

4. Calibration of key components

Detector Wavelength Verification: Verify the wavelength accuracy of the UV detector using a caffeine standard or a holmium oxide filter.

Injection valve test: Manually trigger the sample injection and observe whether the switching is smooth and whether there are any leaks.

Vacuum system recovery: Turn on the mechanical pump, and after the forepump vacuum drops below 100 mTorr, turn on the molecular turbo pump. Evacuate the system for at least 4 hours until the vacuum stabilizes.

Ion source cleaning: Before first use after the holiday, remove the ion source assembly and ultrasonically clean it (methanol:water = 1:1). After thoroughly drying it, reassemble it.

Tuning verification: Run the automatic tuning program and compare the tuning report with the pre-event baseline. If the peak intensity drops by more than 30%, consider the possibility of ion source contamination or detector aging.

(3) General Instrument Restart: Confirm one by one to eliminate potential hazards.

Power-on flush: First, drain any remaining water from the water tank (if it hasn't been drained before the holiday, thoroughly clean it). Then, restart water production and flush the pipelines for 30 minutes.

Water quality verification: Test the resistivity (>18.2 MΩ·cm) and TOC value; only after meeting the standards can it be used.

Stress check: After activation, if the time it takes for the pressure to rise to 0.4–0.6 MPa noticeably increases, you should check the air compressor filter element.

Molecular sieve activation: Run continuously for more than 4 hours to ensure thorough regeneration of the molecular sieve.

3. Centrifuge/Refrigerator/Cultivator

No-load trial run: Perform a no-load run of the centrifuge to check for noise and vibration; only place samples into the refrigerator/incubator after monitoring confirms that the temperature has stabilized at the set value.

Alarm Function Test: Manually trigger the overtemperature alarm and confirm that the SMS/audible and visual alarms are functioning properly.

4. Data Baseline Validation: Ensuring Experimental Comparability 
 

 


 

1. System suitability test

Run the standard sample sequence and calculate the RSD of retention times (should be <1.0%) and the RSD of peak areas (should be <2.0%).

Compared with the data before the holiday, if the deviation exceeds the allowable limit, you need to check the status of the mobile phase, chromatographic column, or detector.

2. Method blank validation

Run the blank solvent to ensure there is no ghost peak interference (if a ghost peak is present, prioritize replacing the ghost peak trapping column).

Run a gradient with a blank gradient to check for baseline drift (a drift of less than 1 mAU/min is considered normal).

3. Re-testing of quality control samples

Select stable quality control samples for injection; only when the results fall within ±3SD on the control chart can the samples be released for testing.
